Project Description

Our purpose is to elucidate, in concrete molecular and cellular terms, the role of mammalian HP1 proteins in parent-of-origin effects in mammals. We have three objectives, which are:
(i) Determine the changes in DNA methylation at imprinted gDMRs in HP1-null TKO ES cells [0-12 months].
(ii) Determine the histone modification changes at imprinted gDMRs in HP1-null TKO ES cells [12-24 months].
(iii) Determine the changes in transcriptional regulation of imprinted genes in HP1-null TKO ES cells [24-36 months]
